Epithalon is a four-amino-acid peptide designed as a simplified copy of a pineal gland extract. The claims around it centre on telomerase, the enzyme that maintains the protective caps on chromosomes. Published work from one institute reports telomerase activation in cell culture and extended lifespan in rodents. That work has not been independently replicated by other groups, which is the single most important thing to know about this compound.
Epithalon is the tetrapeptide Ala-Glu-Asp-Gly (AEDG), derived by sequence reduction from Epithalamin, a bovine pineal gland extract. The proposed mechanism is epigenetic: the peptide is reported to enter the nucleus and bind specific DNA sequences in promoter regions, modulating transcription of genes including TERT. Reported effects include induction of telomerase activity and telomere elongation in cultured human fibroblasts, normalization of melatonin rhythm in aged animals, and restoration of thymic architecture. Independent structural or binding data confirming direct DNA interaction are not available in the mainstream literature, and the reported effects have not been reproduced outside the originating research programme.
