Fat cells contain an enzyme called NNMT that consumes NAD building blocks and helps the cell store energy rather than burn it. 5-Amino-1MQ blocks that enzyme. In obese mice, blocking it raised NAD levels inside fat tissue, shrank fat cells and reduced body weight without changing how much the animals ate. Nothing equivalent has been run in people, and it is worth being clear that this is a quinolinium salt, not a peptide at all.
Nicotinamide N-methyltransferase (NNMT) catalyses methylation of nicotinamide to 1-methylnicotinamide, consuming S-adenosylmethionine and diverting nicotinamide away from NAD+ salvage. 5-Amino-1-methylquinolinium is a cell-permeant bisubstrate-competitive NNMT inhibitor with reported sub-micromolar potency. In diet-induced obese mice, oral dosing increased adipose NAD+ and SAM, reduced adipocyte size and produced weight loss without altered food intake. Selectivity across the broader methyltransferase family, oral bioavailability in humans, and any chronic toxicology package are not established in the peer-reviewed record.
