A nine-amino-acid peptide isolated from rabbit brain in 1977 and named for the sleep pattern it induced. The human sleep evidence has never been convincing.
DSIP was found by taking blood from rabbits in induced sleep and isolating the fraction that put other rabbits to sleep. It was named for that effect. Fifty years later, its receptor has still not been identified, its physiological role is unclear, and human studies of sleep have produced small and inconsistent results. Some of the more interesting human work is not about sleep at all but about opioid and alcohol withdrawal.
A nonapeptide (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) originally isolated from cerebral venous blood of rabbits in electrically induced sleep. No specific receptor has been characterised, and it is not established as a bona fide neurotransmitter or neuromodulator. Reported effects across the literature include modulation of delta-wave EEG activity, altered corticotropin and somatotropin release, and antinociception, with considerable inconsistency between groups. Small human studies in chronic insomnia showed limited effect; more consistent findings come from small trials in opiate and alcohol withdrawal syndromes.

The name is more convincing than the data. The peptide was named for an effect observed in rabbits; controlled human studies with polysomnography showed small and inconsistent changes, and there has been no rigorous modern trial to settle it.
